DAB即二氨基联苯胺(3,3’-diaminobenzidine),辣根过氧化物酶(Peroxidase)最常用的生色底物,其显色原理是在过氧化氢的作用下失去电子而呈现出颜色变化和积累,形成棕色不溶性产物。该反应灵敏度高、特异性好,常用于免疫组化、原位杂交、Western Blot等膜显色或检测细胞或组织内源性的过氧化物酶。由于生成的显色产物不溶于乙醇,因此,在DAB显色后,还可以使用溶于乙醇的染料进行后续染色。可配合DAB染色使用的复染方法包括亚甲基蓝、甲基绿和苏木精。DAB,也称硒试剂,可用于分光光度法检测硒含量。

本品为四盐酸盐形式DAB,易溶于水,可制备成1%储存液,-20℃分装冻存。然后使用前用合适pH的缓冲液(pH7.0~7.6)稀释到工作液,并与过氧化氢(H2O2)工作液混匀,即得到完整的DAB显色液。
