细胞在发生凋亡时,会激活一些DNA内切酶,这些内切酶会切断核小体间的基因组DNA。细胞凋亡时抽提DNA进行电泳检测,可以发现180-200 bp的DNA ladder。
TUNEL (TdT mediated dUTP Nick End Labeling) 细胞凋亡检测试剂盒(YSFluorTM 640)可以用来检测组织细胞在凋亡晚期过程中细胞核DNA的断裂情况。其原理是在末端脱氧核糖核苷酸转移酶(Terminal Deoxynucleotidyl Transferase, TdT)的作用下,在基因组DNA断裂时暴露出的3´-羟基(3´-OH)末端掺入YSFluorTM 640-12-dUTP,从而可以用荧光显微镜或流式细胞仪检测。YSFluorTM 640为红色荧光染料,具有很高的稳定性和很强的亮度。
应用范围广,可以用于检测冷冻或石蜡切片中的细胞凋亡情况,也可以检测培养的贴壁细胞或悬浮细胞的凋亡情况。

Q:血管内皮细胞,想要免疫荧光和 TUNEL 同时进行,不知道该先做哪个? TUNEL 要求立即检测?
A:先做 TUNEL,再进行免疫荧光,这个主要是怕免疫荧光对 TdT 酶产生影响。只要避光保存就可以不立即进行检测。
Q:感觉 DNA 染的有点弥散状,核定位的效果也不是很好,是染色时间太久?感觉细胞质中也像是染到了?
A:有非特异性染色,可能是因为时间太久或者是试剂浓度太高导致的。建议降低染色液浓度或减少染色时间。
