一、SIRP家族概述
SIRP(Signal Regulatory Protein,信号调节蛋白)家族是一类免疫球蛋白超家族跨膜糖蛋白,主要分为三个成员:SIRPα(CD172a)、SIRPβ(CD172b)和SIRPγ(CD172g)。三者具有相似的胞外IgV样结构域,但信号传导机制和功能截然不同。
二、SIRPα(CD172a)- 抑制型受体
2.1 基本特征
- 基因位置:人类染色体20p13
- 结构:胞外3个Ig样结构域(IgV-IgC1-IgC2)+ 跨膜段 + 胞质尾(含2个ITIM基序)
- 表达细胞:髓系细胞(巨噬细胞、树突状细胞、单核细胞)、神经元
- 配体:CD47(高亲和力,Kd~200 nM)
2.2 V1-V8剪接变体
V1-V8代表SIRPα基因的不同剪接变体,由mRNA选择性剪接产生:
